I get this error when trying to open EQMap for the first time on my new system

I'm guessing it has something to do with Vista 64bit and the directories I've got the game installed too. Any help would be appreciated.

That is Vista's user account control UAC security feature

Yeah, its all Vista. Vista doesn't like things messing around inside Program Files without admin rights.
